Changes in the Transcriptome Profiles of Human Amnion-Derived Mesenchymal Stromal/Stem Cells Induced by Three-Dimensional Culture: A Potential Priming Strategy to Improve Their Properties
Alessia Gallo,Nicola Cuscino,Flavia Contino,Matteo Bulati,Mariangela Pampalone,Giandomenico Amico,Giovanni Zito,Claudia Carcione,Claudio Centi,Alessandro Bertani,Pier Giulio Conaldi,Vitale Miceli +11 more

3D Culture and Interferon-γ Priming Modulates Characteristics of Mesenchymal Stromal/Stem Cells by Modifying the Expression of Both Intracellular and Exosomal microRNAs
Matteo Bulati,Alessia Gallo,Giovanni Zito,Rosalia Busà,Gioacchin Iannolo,Nicola Cuscino,Salvatore Castelbuono,Claudia Carcione,Claudio Centi,Gennaro Martucci,Alessandro Bertani,Maria Pia Baiamonte,Cinzia Maria Chinnici,Pier Giulio Conaldi,Vitale Miceli +14 more
TL;DR: It is demonstrated that both 3D culture and IFN-γ treatment are able to modify intracellular and exosomal miRNAs, and the findings might contribute to a better understanding of the molecular mechanisms underlying the miRNA-mediated beneficial effects of MSCs.

Caspase-8 activation by cigarette smoke induces pro-inflammatory cell death of human macrophages exposed to lipopolysaccharide
Marta Cristaldi,Marco Buscetta,Maura Cimino,Agnese La Mensa,Maria Rita Giuffre',Luigi Fiore,Claudia Carcione,Fabio Bucchieri,Francesca Rappa,Claudia Coronnello,Nicolina Sciaraffa,Santina Amato,Tommaso Silvano Aronica,Giovanna Lo Iacono,Alessandro Bertani,Elisabetta Pace,Chiara Cipollina +16 more
TL;DR: The findings show that CSE sensitizes human macrophages to cell death by promoting pyroptotic and apoptotic pathways upon encountering LPS, and it is proposed that while the delayed inflammatory response may result in ineffective defenses against infections, the observed cell death associated with DAMP release may contribute to establish chronic inflammation.

Cigarette smoke promotes inflammasome‐independent activation of caspase‐1 and ‐4 leading to gasdermin D cleavage in human macrophages
Marco Buscetta,Marta Cristaldi,M. Cimino,Agnese La Mensa,Paola Dino,Fabio Bucchieri,Francesca Rappa,Santina Amato,Tommaso Silvano Aronica,Elisabetta Pace,Alessandro Bertani,Chiara Cipollina +11 more
TL;DR: The findings revealed that ASC‐independent activation of caspase‐1, ‐4, and ‐8 and GSDMD cleavage upon exposure to CS may contribute to macrophage dysfunction and feed the chronic inflammation observed in the smokers' lung.

- 10 Mar 2022
TL;DR: In this article , the role of inflammasomes in human primary bronchial epithelial cells (PBECs) exposed to cigarette smoke extract (CSE) was investigated in PBECs isolated from healthy donors.
