Metal-catalyzed cross-coupling reactions

Intracellular membrane traffic defines a complex network of pathways that connects many of the membrane-bound organelles of eukaryotic cells. Although each pathway is governed by its own set of factors, they all contain Rab GTPases that serve as master regulators. In this review, we discuss how Rabs can regulate virtually all steps of membrane traffic from the formation of the transport vesicle at the donor membrane to its fusion at the target membrane. Some of the many regulatory functions performed by Rabs include interacting with diverse effector proteins that select cargo, promoting vesicle movement, and verifying the correct site of fusion. We describe cascade mechanisms that may define directionality in traffic and ensure that different Rabs do not overlap in the pathways that they regulate. Throughout this review we highlight how Rab dysfunction leads to a variety of disease states ranging from infectious diseases to cancer.

The three zones of adrenal cortex are thought to arise from a single multipotential stem cell, but the mechanisms underlying the zonal differentiation during embryonic development of adrenal cortex are poorly understood. Employing subtraction cloning strategy, we isolated three distinct clones that were specifically expressed in the rat glomerulosa zone. One clone, named zona glomerulosa specific clone, encoded a membrane-spanning protein with a signal peptide at the N-terminus, six epidermal growth factor-like repeat motifs, and a transmembrane domain near the C-terminus. It was identified as a rat homolog of preadipocyte factor-1 (Pref-1), a factor involved in maintaining the undifferentiated status of preadipocyte. Immunohistochemical studies confirmed the presence of Pref-1 protein in the glomerulosa zone. Cloning of a membrane-spanning protein with epidermal growth factor- like repeat motifs from adrenal glomerulosa cells

Background. While the clinical validity of Doppler ultrasonography in renal parenchymal disease is still controversial, we have previously reported that the resistive index (RI) and the atrophic index (AI) could estimate tubulointerstitial injury. We aimed to determine whether these indices could estimate renal prognosis in chronic kidney disease (CKD). Methods. We performed a 2-year follow-up study with an observational cohort of 311 CKD patients. The patients were examined by Doppler ultrasonography to calculate RI and AI to be calculated. Glomerular filtration rate (GFR) was estimated with the abbreviated MDRD study equation every 6 months. Results. When we divided the patients into three groups by the RI value of 0.65 and 0.70, there were significant differences in the decrease in GFR among the three groups at 24 months. Kaplan–Meier analysis also showed a significant difference among the three groups in the survival rate of worsening renal function, which was defined as a decrease in GFR of at least 20 mL/min 1.73m 2 or the need for long-term dialysis therapy until the end of the 2-year follow-up. Cox proportional-hazard analysis identifiedovertproteinuria(≥1.0g/gcreatinine),highRI(>0.70) and high systolic blood pressure (≥140 mmHg) as independent predictors of worsening renal function. In contrast, AI was of no significance in evaluating renal prognosis in CKD. Conclusions. This study suggested that RI, and proteinuria and hypertension were independent risk factors for the progression of CKD.

AIM: To examine the effects of adiponectin on the functions of Kupffer cells, key modulators of lipopolysaccharide (LPS) -induced liver injury. METHODS: D-galactosamine (GalN) and LPS were injected intraperitoneally into adiponectin-/- mice and wild type mice. Kupffer cells, isolated from Sprague-Dawley rats, were preincubated with or without adiponectin, and then treated with LPS. RESULTS: In knockout mice, GalN/LPS injection significantly lowered the survival rate, significantly raised the plasma levels of alanine transaminase and tumor necrosis factor-α (TNF-α)signifi cantly reduced IL-10 levels compared with wild type mice. TNF-α gene expression in the liver was which higher and those of IL-10 were lower in knockout mice than in wild type mice. In cultured adiponectin-pre-treated Kupffer cells, LPS signifi cantly lowered TNF-α levels and raised IL-10 levels in the culture media and their respective gene expression levels, compared with Kupffer cells without adiponectinpre-treatment.
