Plant-specific SQUAMOSA promoter-binding protein-like (SPL) transcription factors play important regulatory roles during plant growth and development, fruit ripening, inflorescence branching, and biotic and abiotic stresses. However, there have been no identification or systematic studies of the SPL gene family in the sweet cherry. In this study, 12 SPL genes were identified in the sweet cherry reference genome, which were distributed over 6 chromosomes and classified into six groups according to phylogenetic relationships with other SPL gene families. Nine PavSPLs were highly expressed at green fruit stages and dramatically decreased at the onset of fruit ripening, which implied that they were important regulators during fruit development and ripening. The expression patterns of PavSPL genes under ABA, GA, and MeJA treatments showed that the PavSPLs were involved in the process of fruit ripening. A subcellular localization experiment proved that PavSPL4 and PavSPL7 proteins were localized in the nucleus. The genome-wide identification of the SPL gene family provided new insights while establishing an important foundation for sweet cherry studies.

Microbial synthesis of cordycepin, current systems and future perspectives

Abstract Proteases help plants maintain protein quality and break down protein subsets in reply to environmental, developmental, biotic, and abiotic stressors. Filamentation temperature-sensitive H (FtsH) is an ATP-dependent metalloprotease detected in both prokaryotes and eukaryotes cells. The present research, the FtsH gene in the bean, which has an important place in the legume family and is an important agricultural product, was characterized for the first time using various bioinformatic tools, and qRT-PCR measured its expression level. In the study, effectively identified and characterized 17 FtsH genes present in genome the of Phaseolus vulgaris . The MW of FtsH proteins varied from 71.16 to 147.07 kDa, their amino acid lengths ranged from 642 to 1284, and their pI values varied from 5.39 to 9.60. Interestingly, the distribution of these 17 distinct PvFtsH genes across the 8 chromosomes was not uniform, exhibiting an uneven pattern throughout the genome. A pair of segmental duplication fragments were found, revealing probable processes of gene expansion and evolution. Collinearity with related genes in Arabidopsis and rice was thoroughly examined to determine the evolutionary conservation and differentiation of PvFtsH genes. Additionally, we used RNAseq and qRT-PCR to investigate the expression patterns of PvFtsH in leaf tissue under salt and drought conditions. Our data showed unique expression patterns, suggesting PvFtsH may respond to environmental and physiological stressors. Overall, this work makes major contributions to our understanding of PvFtsH genes and their roles in the context of gene evolution, chromosomal distribution, and expression patterns under various environmental situations.

ABSTRACT In the past two decades, promoter engineering has been extensively employed as a powerful strategy for fine‐tuning the regulation of gene transcription to awaken or increase the expression of secondary metabolite (SM) biosynthetic gene clusters (BGCs) in fungal genomes. This comprehensive review provides an overview of recent advances in various fungal promoter engineering techniques, including promoter replacement, optimization, and combinatorial methods, and their application in the enhancement of SM production. Additionally, current challenges and future prospects for efficient promoter engineering for precise metabolic regulation in fungi are discussed.

Genome-wide identification of cysteine-rich receptor-like kinases in sweet cherry reveals that PaCRK1 enhances sweet cherry resistance to salt stress

Abstract Background Cordyceps militaris , a kind of edible and medicinal fungus widely accepted in East Asia, has attracted much attention as a potential cell factory for producing adenosine analogs. Despite the rapid development in gene editing techniques and genome modeling, the diversity of DNA elements in C. militaris was too short to achieve rational heterogeneous expression for metabolic engineering studies. Results In this study, P trpC , a kind of promoter with a relatively appropriate expression level and small size, was selected as a monomer for promoter library construction. Through in vitro BioBricks assembly, 9 overlapping P trpC promoters with different copy numbers as well as reporter gene gfp were connected and subsequently integrated into the genome of C. militaris . Both the mRNA transcription level and the expression level of gene gfp gradually increased along with the copy number of the overlapping promoter NP trpC and peaked at 7. In the meantime, no significant difference was found in either the biomass or morphological characteristic of engineered and wild-type strains . Conclusions This study firstly expanded the overlapping promoter strategy used in model microorganism in C. militari s. It was a proof-of-concept in fungi synthetic biology and provide a general method to pushed the boundary of promoter engineering in edible mushroom.
