INTRODUCTION The presence of Reed-Sternberg malignant cells is absolutely necessary for Hodgkin's lymphoma diagnostic, but it is not always sufficient because can be observed Reed-Sternberg-like cells in other malignant and benign diseases, too. The CD30 expression at Hodgkin and Reed-Sternberg level can give us supplementary information in differential diagnostic and can be used as progressive disease factor. MATERIAL AND METHODS Our study was composed from 63 cases histopathological diagnosed with Hodgkin's lymphoma and hospitalized in Hematology Department of County Hospital Timisoara. CD30 expression was immunohistochemical semi-quantitative evaluated using clone BerH2 as primary antibody and APAAP-New Fuchsin as visualization system. RESULTS AND DISCUSSIONS The increasing of CD30 expression occurs in the same time with advanced stages and the disease progression (p =0.001). For I and II stages CD30 expression does not overcome (-/+) category while the III and IV stages, all the cases are situated in (+/-) and (+) categories. No connection can be noticed between histological type and CD30 expression (p < or = 1). We consider that using this staining, although less used in Romania, must be done in all Hodgkin's lymphoma and Hodgkin's lymphoma-like cases. We say that because the main cause of relapses is represented by inadequate clinical staging and diagnostic. CONCLUSIONS In our study, the increasing of CD30 expression is associated with advanced disease stage. We recommend reinvestigating and restaging all cases that was included into an incipient stages and they have a CD30 expression situated in (+/-) and (+) intervals because some lymph nodes could be overlooked.

CD30 expression utilization for the accuracy of classical Hodgkin's lymphoma staging.

More than 40% patient of Hodgkin lymphoma (HL) are chemoresistance and show early disease relapse that leads to poor prognostic. Epstein-Barr Virus (EBV) appears to have poor prognostic in HL. The CD30 expression at Hodgkin and Reed-Stenberg cells suggested has associated with advanced disease stage. The International Prognostic Score-7 (IPS) is a seven-parameter of risk stratification index for prognostic and as a consideration for treatment plan. The aim of this journal is to see correlation between EBV infection and CD30 expression with IPS-7 in Classic HL (CHL) patient.
Case-control research design was done to 40 Fixed Formalin Paraffin-Embedded (FFPE) of CHL at the Dr Hasan Sadikin Hospital, Bandung. Cases were 20 patients CHL of high-risk score IPS-7 and controls were 20 patients with low-risk score. Detection of EBV-encoded small RNA-1 (EBER1) by Real-Time RT-PCR and Immunohistochemical staining of CD30 was performed to all samples. All data were analyzed using Chi-Square test with p-value <0.05 of significant level.
EBV was present in 10 cases of CHL (25%). The result of this study shows a different statistically significant of EBV infection (p=0.028) and CD30 expression (p=0.004) between low risk and high-risk IPS-7 HL. CD30 expression (OR= 7.429) may provide stronger factor for risk assessment for the patient with HL, compare to EBER1 detection (OR=6.000).
This study concluded that CD30 expression and EBV Infection correlate with risk assessment IPS-7 in CHL.

CD30 expression and Detection of Epstein-Barr Virus-Encoded Small RNA by Real-Time RT-PCR of Classic Hodgkin Lymphoma and Their Correlation with Risk Stratification

Introduction and Aim: Nasopharyngeal cancer (NPC) prognosis has been associated with cyclin D1 and Ki-67. These tumor markers had never been tested in NPC patients in Indonesia before. Therefore, this study aims to evaluate the relationships between Ki-67 and cyclin D1 expression levels and the clinicopathological features of nasopharyngeal cancer (NPC) patients.
 
Materials and Methods: During the years 2015 until 2018, thirty-one nasopharyngeal cancer patients were studied in a cohort study at RSUPN Dr. Cipto Mangunkusumo. Patients aged 18 years, diagnosed with NPC according to the histopathological results, metastasis NPC, Eastern Cooperative Oncology Group 0–1, and those who had completed first-line chemotherapy treatment with 5–fluorouracil were included in this study. The Ki-67 and cyclin D1 expression levels were evaluated with the h-score. The NLR values were collected prior to the treatment. The 2-year survival rate was analyzed based on the optimal cut-off value obtained. The optimal cut-off values of Ki-67 and cyclin D1 were categorized into high- and low-expression groups to observe the 2-year mortality.
 
Results: The relationships between Ki-67 and cyclin D1, clinical, pathological, and prognosis of patients were examined. Patients with Ki-67 high expression had a worse 2-year survival rate than those with low expression (p0.05). Ki-67 expression was found to be significantly related to adverse outcomes (p0.05, OR: 41.107, 95% CI: 2.600–649.940), while the cyclin D1 expression was not significantly related to adverse outcomes (p0.05).
 
Conclusion: Ki-67 expression was associated with poor prognosis in NPC patients. Nevertheless, further research is required to explore the specific mechanism of cyclin D1 expression in NPC prognosis.

Expression levels of Ki-67 and cyclin D1 in advanced nasopharyngeal carcinoma and its prognosis

BACKGROUND: Dyspareunia and pain due to the decrease of vaginal wall thickness usually happen in menopausal women. The reduction of estrogen levels cause the decreasing of cell proliferation and the thinning of vaginal wall epithelium. Tomato ( Solanum lycopersicum ) is a source of phytoestrogens, which produce estrogenic effects. This study aims to assess the effect of tomato juice on Ki-67 expression and epithelial thickness of the vaginal wall in menopausal rats. METHODS: This was an experimental study using Sprague-Dawley rats. Twenty-four female rats, aged 4 months and weighing 150-200 grams, were divided into 4 groups. Each group consisted of 6 rats. Negative control (NC) group was group of rats with sham procedure and performed by distilled water for 28 days. Positive control (PC) group was group of bilateral ovariectomized rats and performed by distilled water for 28 days. The first treatment (T1) group was group of bilateral ovariectomized rats, given tomato juice at dose of 11g/200g body weight (BW)/day. The second treatment (T2) group was group of bilateral ovariectomized rats, given tomato juice at dose of 15g/200g BW/day. Data analysis was done with Anova, multiple comparisons and regression test. RESULTS: The group with lowest Ki-67 expression was PC group (2.52±0.60). The expression of Ki-67 in treatment groups (T1 and T2) was higher than PC group, but lower than NC group. There was no significant difference between groups ( p =0.771). The lowest epithelial thickness was found in PC group (21.19±3.96) and the highest was found in the treatment groups (38.73±12.43). There was positive correlation between tomato juice and epithelial thickness ( p =0.647). CONCLUSION: Tomato juice increases Ki-67 expression and epithelial thickness on the vaginal wall of menopausal rats. The increase of epithelial thickness follows the administration dose, but Ki-67 expression does not exceed in the control rats. Tomato juice increases the cell proliferation of vaginal wall on menopausal rats, however the increase is still within normal limits. KEYWORDS: epithelial thickness, Ki-67, menopause, tomato juice

The Effect of Tomato Juice in Increasing Ki-67 Expression and Epithelial Thickness on The Vaginal Wall of Menopausal Rats

Liposarkoma merupakan salah satu tumor ganas jaringan lunak dan terbanyak ditemukan pada usia dewasa antara dekade 5 dan 7 serta jarang terjadi pada usia anak. Liposarkoma sering terjadi pada ekstremitas bawah (fosa poplitea dan paha bagian tengah), retroperitoneal, perirenal, mesenteric region, dan area bahu. Penelitian ini bersifat retrospektif dari 30 kasus liposarkoma yang diambil dari data Departemen Patologi Anatomi Rumah Sakit Dr. Hasan Sadikin Bandung yang terdiri atas 20 kasus low-grade liposarcoma dan 10 kasus high-grade liposarcoma. Penelitian dilakukan periode Februari–Mei 2011. Dilakukan pemeriksaan khusus imunohistokimia dengan menggunakan antibodi monoklonal Ki-67 yang berfungsi sebagai marker proliferasi sel, dengan tujuan melihat ekspresi Ki-67 dan apakah terdapat hubungan antara ekspresi Ki-67 dan grading histopatologi liposarkoma. Ekspresi dan intensitas warna Ki-67 pada sel tumor dibagi menjadi sangat rendah ( 80% positif), sedangkan untuk intensitas warna Ki-67 dibagi menjadi lemah, sedang, dan kuat. Terdapat perbedaan yang signifikan antara ekspresi dan intensitas Ki-67 pada low-grade liposarcoma dan high-grade liposarcoma (p 80% positive), whereas the intensity of Ki-67 staining was into weak, moderate and strong. There were significant differences between the Ki-67 expression and intensity level according to the histopathological types of liposarcoma (p<0.0001). In conclusions, Ki-67 expression increases with strong intensity of Ki-67 only in high-grade liposarcoma, so relates with progressivity of tumor. Hubungan Ekspresi Ki-67 dengan Grading Histopatologi Liposarkoma

Nasopharyngeal carcinoma (NPC) has a high incidence and mortality rate in Southeast Asia included in Indonesia. Radioresistance is a major obstacle for a successful treatment of NPC. DNA repair in the cell cycle and angiogenesis factors affect the response of tumor cells to radiotherapy. Cyclin D1 that functions in the cell cycle process and vascular endothelial growth factor (VEGF) as an angiogenesis factor are considered to play a role in the occurrence of radioresistance. The objective of this study was to evaluate the association between cyclin D1 and VEGF expressions with radiotherapy response in undifferentiated NPC. This study used a retrospective case control analysis design. Secondary data from medical records of patients diagnosed as undifferentiated NPC who received a complete radiotherapy at the Department of Radiation Oncology, Dr. Hasan Sadikin General Hospital, Bandung. There were 44 samples divided into radiosensitive (22 samples) and radioresistant (22 samples) groups. Immunohistochemical examination of cyclin D1 and VEGF expressions was performed on paraffin blocks of patients’ nasopharyngeal biopsy. Data analysis used Chi-Square test with p ≤0.05. Cyclin D1 was expressed strongly in 86.4% of the radioresistant group and 59.1% in the radiosensitive group (p 0.05). In conclusion, there is significant association between cyclin D1 expression with radiotherapy response in undifferentiated NPC. However, there is no association between VEGF expression with radiotherapy response.

The role of cyclin D1 and vascular endothelial growth factor (VEGF) in radiotherapy response of undifferentiated nasopharyngeal carcinoma

Nasopharyngeal carcinoma ( N PC) is an epithelial malignancy originating from the surface of the lateral and posterior wall of the nasopharynx. Bcl-2 is an oncoprotein that plays an important role in disrupting the process of apoptosis. Expression of Bcl-2 in biopsy samples of undifferentiated NPC also related to the nature of more aggressive tumor mass and unfavorable clinical radiotherapy response. The purpose of this study was to assess the relationship of Bcl-2 expression with radiotherapy response in undifferentiated NPC. The method of this study areusing 40 samples of paraffin blocks were diagnosed as undifferentiated NPC by histopathologic examination with H&E staining. The samples were divided into 2 groups who respond and do not respond to radiotherapy. All samples examined by Bcl-2 immunohistochemistry and the expression willanalyzed and assess therelationship to radiotherapy response. Of the 40 cases of undifferentiated NPC show imunoekspresi Bcl-2 positive in 39 cases (97.5%) and 1 case (2.5%) negative from response group. The results of statistical analysis of the Bcl-2 obtained significant results with p value <0.05 so we can conclude there is a relationship between Bcl-2 expression with radiotherapy response. Conclusion, Bcl-2 can be used as a predictor for the success of radiotherapy in undifferentiated NPC. Keywords: Bcl-2, undifferentiated NPC, radiotherapy response

Bcl-2 Immunoexpression as Radiotherapy Response Predictor in Undifferentiated Nasopharynx Carcinoma

BackgroundSarcomas are rare, aggressive tumors originating from mesenchymal tissue. Radiotherapy is currently accepted as thestandard adjuvant to the treatment of high-grade sarcomas, whereas low-grade sarcomas are controversial, but the response ofradiotherapy to sarcomas is up to 50%. The aim of this study was to determine the association between the histopathologicalgrade of sarcoma and the radiotherapeutical response.MethodsThe research design was a cross-sectional study, with retrospective analysis. The sample consisted of 80 paraffin block tissuesfrom biopsy and surgery specimens of sarcoma from January 2013 to December 2019 at Hasan Sadikin Hospital in Bandung,consisting of 45 radiosensitive samples and 35 radioresistant samples. All samples were assessed for histopathological gradeaccording to the FNCLCC. Statistical analysis used the Chi-Square test with a significant value of p <0.05. Data were processedusing SPSS version 24.0 for windows.ResultsIn the radiosensitive group, 15 (33.3%) samples were low grade sarcomas and 30 (66.7%) samples were high grade sarcoma.There was no statistically significant association between histopathological grade and radiotherapeutical response in sarcomasp> 0.05 (p = 0.304).ConclusionIn this study, the radiotherapeutical response was not associated to the histopathological grade of sarcoma. This is because ofthe sensitivity of radiotherapy is influenced by other factors including molecular characteristics.
