Small molecule obatoclax (GX15-070) antagonizes MCL-1 and overcomes MCL-1-mediated resistance to apoptosis
Mai Nguyen,Richard C. Marcellus,Anne Roulston,Mark F. Watson,Lucile Serfass,S.R. Murthy Madiraju,Daniel Goulet,Jean Viallet,Laurent Bélec,Xavier Billot,Stephane Acoca,Enrico O. Purisima,Adrian Wiegmans,Leonie A. Cluse,Ricky W. Johnstone,Pierre Beauparlant,Gordon C. Shore +16 more
TL;DR: Findings support a rational clinical development opportunity for the compound in cancer indications or treatments where MCL-1 contributes to resistance to cell killing.

Analysis of the apoptotic and therapeutic activities of histone deacetylase inhibitors by using a mouse model of B cell lymphoma
Ralph K. Lindemann,Andrea Newbold,K. Whitecross,Leonie A. Cluse,Ailsa J. Frew,Leigh Ellis,Steven P. Williams,Adrian Wiegmans,Anthony E. Dear,Clare L. Scott,Marc Pellegrini,Andrew H. Wei,Victoria M. Richon,Paul A. Marks,Scott W. Lowe,Mark J. Smyth,Ricky W. Johnstone +16 more
TL;DR: Vorinostat demonstrated a direct correlation between induction of tumor cell apoptosis in vivo and therapeutic efficacy and have broad implications regarding stratification of patients receiving HDACi therapy alone or in combination with other anticancer agents.

Defining the target specificity of ABT-737 and synergistic antitumor activities in combination with histone deacetylase inhibitors.
K. Whitecross,Amber E. Alsop,Leonie A. Cluse,Adrian Wiegmans,Kellie-Marie Banks,Claudia P. Coomans,Melissa J. Peart,Andrea Newbold,Ralph K. Lindemann,Ricky W. Johnstone,Ricky W. Johnstone +10 more
TL;DR: Emu-myc lymphomas that expressed Bcl-2 throughout the tumorigenesis process were especially sensitive to ABT-737, while those forced to overexpress Mcl-1 were not, which supports the notion that tumor cells "addicted" toABT- 737 target proteins are likely to be the most sensitive target cell population.

Deciphering the Molecular Events Necessary for Synergistic Tumor Cell Apoptosis Mediated by the Histone Deacetylase Inhibitor Vorinostat and the BH3 Mimetic ABT-737
Adrian Wiegmans,Amber E. Alsop,Michael Bots,Leonie A. Cluse,Steven P. Williams,Kellie-Marie Banks,Rachael A Ralli,Clare L. Scott,Anna Frenzel,Andreas Villunger,Ricky W. Johnstone +10 more
TL;DR: The apoptotic proteins and pathways necessary for synergistic tumor cell apoptosis and in vivo antitumor responses seen when the HDAC inhibitor vorinostat is combined with the BH3-mimetic ABT-737 in lymphomas overexpressing Bcl-2 are identified.
