Nanocarbons with different dimensions (e.g., 0D fullerenes and carbon nanodots, 1D carbon nanotubes and graphene nanoribbons, 2D graphene and graphene oxides, and 3D nanodiamonds) have attracted enormous interest for applications ranging from electronics, optoelectronics, and photovoltaics to sensing, bioimaging, and therapeutics due to their unique physical and chemical properties. Among them, nanocarbon-based theranostics (i.e., therapeutics and diagnostics) is one of the most intensively studied applications, as these nanocarbon materials serve as excellent biosensors, versatile drug/gene carriers for specific targeting in vivo, effective photothermal nanoagents for cancer therapy, and promising fluorescent nanolabels for cell and tissue imaging. This review provides a systematic overview of the latest theranostic applications of nanocarbon materials with a comprehensive comparison of the characteristics of different nanocarbon materials and their influences on theranostic applications. We first introduce the different carbon allotropes that can be used for theranostic applications with their respective preparation and surface functionalization approaches as well as their physical and chemical properties. Theranostic applications are described separately for both in vitro and in vivo systems by highlighting the protocols and the studied biosystems, followed by the toxicity and biodegradability implications. Finally, this review outlines the design considerations for nanocarbon materials as the key unifying themes that will serve as a foundational first principle for researchers to study, investigate, and generate effective, biocompatible, and nontoxic nanocarbon materials-based models for cancer theranostics applications. Finally, we summarize the review with an outlook on the challenges and novel theranostic protocols using nanocarbon materials for hard-to-treat cancers and other diseases. This review intends to present a comprehensive guideline for researchers in nanotechnology and biomedicine on the selection strategy of nanocarbon materials according to their specific requirements.

Nanocarbons for Biology and Medicine: Sensing, Imaging, and Drug Delivery

As a class of carbon-based nanomaterials, carbon dots (CDs) have attracted enormous attention because of their tunable optical and physicochemical properties, such as absorptivity and photoluminescence from ultraviolet to near-infrared, high photostability, biocompatibility, and aqueous dispersity. These characteristics make CDs a promising alternative photonic nanoagent to conventional fluorophores in disease diagnosis, treatment, and healthcare managements. This review describes the fundamental photophysical properties of CDs and highlights their recent applications to bioimaging, photomedicine (e.g., photodynamic/photothermal therapies), biosensors, and healthcare devices. We discuss current challenges and future prospects of photonic CDs to give an insight into developing vibrant fields of CD-based biomedicine and healthcare.

Photonic Carbon Dots as an Emerging Nanoagent for Biomedical and Healthcare Applications.

Abstract Multi-color fluorescent nanodiamonds (FNDs) containing a variety of color centers are promising fluorescent markers for biomedical applications. Compared to colloidal quantum dots and organic dyes, FNDs have the advantage of lower toxicity, exceptional chemical stability, and better photostability. They can be surface functionalized by techniques similar to those used for other nanoparticles. They exhibit a variety of emission wavelengths from visible to near infrared, with narrow or broad bandwidths depending on their color centers. In addition, some color centers can detect changes in magnetic fields, electric fields, and temperature. In this article review, we will discuss the current trends in FND’s development, including comparison to the early development of quantum dots. We will also highlight some of the latest advances in fabrication, as well as demonstrations of their use in bioimaging and biosensing.

A great challenge in noninvasive biomedical imaging is the acquisition of images inside a biological system at the cellular level Common modalities used today such as magnetic resonance or computed tomography imaging have the advantage that any part of a living organism can be imaged at any depth, but are limited to millimeter resolution and can usually not be employed eg, for surgical guidance Optical imaging techniques offer resolution on the 100 nanometer scale, but are limited by the strong attenuation of visible light by biological matter and are traditionally used to image on the surface Near-infrared light in the "biological windows" can penetrate much deeper into biological samples, rendering fluorescence-based imaging a viable alternative In the past two decades, many fluorescent nanomaterials have been developed to operate in the near infrared, yet only few materials emitting above 1000 nm exist and none are approved for clinical use This review describes recent advances in the development and use of near-infrared fluorescent nanomaterials for biomedical imaging and sensing applications The physical and chemical properties as well as the bioconjugation and application of materials such as organic fluorophores, semiconductor quantum dots, carbon-based materials, rare earth materials, and polymer particles are discussed

Near-Infrared Fluorescent Nanomaterials for Bioimaging and Sensing

Diamond particles containing color centers—fluorescent crystallographic defects embedded within the diamond lattice—outperform other classes of fluorophores by providing a combination of unmatched photostability, intriguing coupled magneto-optical properties, intrinsic biocompatibility, and outstanding mechanical and chemical robustness. This exceptional combination of properties positions fluorescent diamond particles as unique fluorophores with emerging applications in a variety of fields, including bioimaging, ultrasensitive metrology at the nanoscale, fluorescent tags in industrial applications, and even potentially as magnetic resonance imaging contrast agents. However, production of fluorescent nanodiamond (FND) is nontrivial, since it requires irradiation with high-energy particles to displace carbon atoms and create vacancies—a primary constituent in the majority color centers. In this review, centrally focused on material developments, major steps of FND production are discussed with emphasis on current challenges in the field and possible solutions. The authors demonstrate how the combination of fluorescent spectroscopy and electron paramagnetic resonance provides valuable insight into the types of radiation-induced defects formed and their evolution upon thermal annealing, thereby guiding FND performance optimization. A recent breakthrough process allowing for production of fluorescent diamond particles with vibrant blue, green, and red fluorescence is also discussed. Finally, the authors conclude with demonstrations of a few FND applications in the life science arena and in industry.

Review Article: Synthesis, properties, and applications of fluorescent diamond particles.

Many novel fluorescent nanomaterials exhibit radically different optical properties compared to organic fluorophores that are still the most extensively used class of fluorophores in biology today. Assessing the practical impact of these optical differences for bioimaging experiments is challenging due to a lack of published quantitative benchmarking data. This study therefore directly and quantitatively compares the brightness and photostability of representatives from seven classes of fluorescent materials in spectroscopy and fluorescence microscopy experiments for the first time. These material classes are: organic dyes, semiconductor quantum dots, fluorescent beads, carbon dots, gold nanoclusters, nanodiamonds, and nanorubies. The relative brightness of each material is determined and the minimum material concentrations required to generate sufficient contrast in a fluorescence microscopy image are assessed. The influence of optical filters used for imaging is also discussed and suitable filter combinations are identified. The photostability of all materials is determined under typical imaging conditions and the number of images that can be acquired is inferred. The results are expected to facilitate the transition of novel fluorescent materials from physics and chemistry into biology laboratories.
