The lion's share of bacteria in various environments cannot be cloned in the laboratory and thus cannot be sequenced using existing technologies. A major goal of single-cell genomics is to complement gene-centric metagenomic data with whole-genome assemblies of uncultivated organisms. Assembly of single-cell data is challenging because of highly non-uniform read coverage as well as elevated levels of sequencing errors and chimeric reads. We describe SPAdes, a new assembler for both single-cell and standard (multicell) assembly, and demonstrate that it improves on the recently released E+V-SC assembler (specialized for single-cell data) and on popular assemblers Velvet and SoapDeNovo (for multicell data). SPAdes generates single-cell assemblies, providing information about genomes of uncultivatable bacteria that vastly exceeds what may be obtained via traditional metagenomics studies. SPAdes is available online ( http://bioinf.spbau.ru/spades ). It is distributed as open source software.

A comprehensive understanding on how plants respond to salinity stress at different levels and an integrated approach of combining molecular tools with physiological and biochemical techniques are imperative for the development of salt-tolerant varieties of plants in salt-affected areas. Recent research has identified various adaptive responses to salinity stress at molecular, cellular, metabolic, and physiological levels, although mechanisms underlying salinity tolerance are far from being completely understood. This paper provides a comprehensive review of major research advances on biochemical, physiological, and molecular mechanisms regulating plant adaptation and tolerance to salinity stress.

Mechanism of Salinity Tolerance in Plants: Physiological, Biochemical, and Molecular Characterization

In present study we focused on the weed occurrence and diversity in rice field according to methods of weed control. Experiments comprised of no herbicide treatment (NHT), herbicide treatment (HT) and golden apple snail treatment (GAST) within the three months (July to September) of growing rice. According to results, five to seven different weed species were found in NHT, while two to four different weed species were found in HT and GAST. Monochoria vaginalis was dominant species during growing period in NHT and HT while Lemna paucicostata was the dominant in July but Persicaria hydropiper was dominant for August and September. Simpson's dominance index was highest in GAST(0.96) compared with other treatments (NHT 0.27 and HT : 0.51). The similarity coefficient was 50.31%, 4.65% and 0.38% for NHT versus HT, NHT versus GAST and HT versus GAST, respectively. In conclusion, the weed species diversity, dominance and similarity coefficient were varied in different weed control methods of rice growing.

Fuelwood is an important source of energy in the Hindukush region of northern Pakistan, as firewood is extensively used for cooking and heating purposes in this remote mountainous area. In current study, we recorded data pertinent to fuelwood consumption and its impact on deforestation in Utror and Gabral valleys, located in the upper Swat valley of Pakistan. It was observed that 27 plant species were utilized as fuelwood species in the project area. However, Cedrus deodara, Quercus dilatata and Pinus willichiana were the most exploited fuelwood plant species. We also observed that annually about 4809.86 tons of valuable wood is used for cooking and heating houses. During summer season, pressure on the forest for fuelwood is comparatively lesser as firewood is not used for heating houses during this period.

Wood as a fuel source in the Hindukush: a case study of Utror and Gabral valleys, northren Pakistan.

The isolation and structure elucidation of a new triterpenic acid named floccosic acid (1) is reported on the basis of the 1D- and 2D-NMR assignments. This secondary metabolite was isolated as a new constituent, along with the known triterpenoids, betulinic acid and β-amyrin. All these compounds were purified by repeated column chromatography of the MeOH extract of Nepeta floccosa. The structure elucidation of the new compound was accomplished by the combined mass spectrometry (MS), infrared (IR) and ultraviolet (UV) absorption spectroscopy, one- (1H- and 13C-) and two-dimensional (HC correlations; HMBC and HSQC) NMR techniques. The known compounds were identified by comparison of their physical and spectroscopic data with those reported in the literature.

A new flavonoid glycoside, named desmiflavanoside (1), was isolated from the MeOH extract of Desmidorchis flava (N.E.Br.) Meve & Liede, and its structure was elucidated based on 1D and 2D NMR spectroscopic techniques as well as mass spectrometry. Preliminary studies demonstrated that although compound 1 is moderately cytotoxic at lower concentrations, its activity plateaus at higher concentrations. Notably, compound 1 demonstrated a significant dose-dependent response to lipid peroxidation and that its antilipid peroxidation was similar (89.6 ± 0.81%) to the known standard (BHT; 91.3 ± 0.49%; p < 0.051). On the other hand, the flavone glycoside 1 did not show inhibitory effects against urease and acetylcholinesterase enzymes.
