Targeting the PD-1/PD-L1 Immune Evasion Axis With DNA Aptamers as a Novel Therapeutic Strategy for the Treatment of Disseminated Cancers
Aaron Prodeus,Aws Abdul-Wahid,Nicholas W. Fischer,Nicholas W. Fischer,Eric Huang,Marzena Cydzik,Jean Gariépy +6 more
TL;DR: The development of DNA aptamers as synthetic, nonimmunogenic antibody mimics, which bind specifically to the murine extracellular domain of PD-1 and block the PD- 1:PD-L1 interaction are reported.

Enhancing Checkpoint Inhibitor Therapy with Ultrasound Stimulated Microbubbles.
TL;DR: Investigation of "anti-vascular" ultrasound-stimulated microbubble (USMB) treatments in combination with anti-PD-1 CI therapy indicates the ability of anti- vascular USMBs to increase the anti-tumor effects of CI therapy; the specific mechanisms of enhancement remain to be established.

Induction of antigen-specific TH 9 immunity accompanied by mast cell activation blocks tumor cell engraftment.
Aws Abdul-Wahid,Aws Abdul-Wahid,Marzena Cydzik,Aaron Prodeus,Aaron Prodeus,Mays Alwash,Mays Alwash,Mile Stanojcic,Megan Thompson,Eric Huang,John E. Shively,Scott D. Gray-Owen,Jean Gariépy,Jean Gariépy +13 more
TL;DR: Vaccination with a recombinant CEA IgV‐like N domain, formulated with the TLR3 ligand poly I:C, elicits a CEA‐specific TH9 response, wherein IL‐9 secreting TH cells act in concert with CEA N domain‐specific antibodies as well as activated mast cells in preventing tumor cell engraftment.

VISTA.COMP - an engineered checkpoint receptor agonist that potently suppresses T cell-mediated immune responses.
Aaron Prodeus,Aws Abdul-Wahid,Amanda Sparkes,Nicholas W. Fischer,Marzena Cydzik,Nicholas Chiang,Mays Alwash,Alessandra Ferzoco,Nathalie Vacaresse,Michael Julius,Reginald M. Gorczysnki,Jean Gariépy +11 more
TL;DR: A stable pentameric VISTA construct is designed by genetically fusing its IgV domain to the pentamerization domain from the cartilage oligomeric matrix protein (COMP) and it is believed that VISTA.COMP is a checkpoint receptor agonist and the first agent to the authors' knowledge targeting the putative VISTA-receptor to suppress T cell-mediated immune responses.

VEGF-B prevents excessive angiogenesis by inhibiting FGF2/FGFR1 pathway
Chun-Chaing Lee,Rongyuan Chen,G. Sun,Xialin Liu,Xianchai Lin,Changying He,Liying Xing,Lixian Liu,Lasse D. Jensen,Anil Kumar,Harald F. Langer,Xiangrong Ren,Jianing Zhang,Lijuan Huang,Xiangke Yin,JongKyong Kim,Juanhua Zhu,Guanqun Huang,Jiani Li,Weiwei Lu,Juanxi Liu,Jiaxin Hu,Qihang Sun,Weisi Lu,Lekun Fang,Shasha Wang,Haiqing Kuang,Yihan Zhang,Geng Tian,Jia Mi,Bibo Kang,Masashi Narazaki,Aaron Prodeus,Luc Schoonjans,David M. Ornitz,Jean Gariépy,Guy Eelen,Mieke Dewerchin,Yunlong Yang,Jingsong Ou,Antonio Mora,Jin Yao,Chen Zhao,Yizhi Liu,Peter Carmeliet,Yihai Cao,Xuri Li +46 more
TL;DR: This work reveals for the first time an unexpected and surprising function of VEGF-B as an endogenous inhibitor of angiogenesis by inhibiting the FGF2/FGFR1 pathway when the latter is abundantly expressed.
