Potential and future scope of nanoemulgel formulation for topical delivery of lipophilic drugs

The paper reviews an overview of a conventional and novel approach in the topical drug delivery system. Drug delivery via the skin is becoming progressively popular due to its convenience and affordability. The skin is the most important mechanical barrier to the penetration of many drug substances and acts as an ideal site to deliver the drug both locally and systemically. The topical route has been a favored route of drug administration over the last decades. Despite conventional topical drug delivery systems limits in poor retention and low bioavailability. This drawback overcomes by extensive research to develop a novel topical drug delivery systems targeting to improve the safety, efficacy and to minimize side effects. The conventional review focuses on dusting powders, poultices, plasters, lotion, liniments, solution, emulsion, suspension, colloidions, tinctures, creams, gels, ointments, pastes, suppositories, transdermal delivery systems, tapes, and gauzes and rubbing alcohol while the novel review focuses on novel gels, aerosol foams, microsponges, muco-adhesive bio-adhesives, novel vesicular carriers, nano-emulsion & nano-emulgel, protein and peptide delivery, polymers, emulsifier-free formulations and fullerenes etc. The key purpose of a topical delivery system is to enhance the skin permeability and to retain in the dermis. This review addresses a basis for further advancement and up-gradation of current techniques and technologies.

Vaginal drug administration can improve prophylaxis and treatment of many conditions affecting the female reproductive tract, which includes fungal and bacterial infections, sexually transmitted diseases and cancer also. This is the best route for the administration of proteins, peptides, and also other therapeutic drugs like macro-molecules. For the administration of drugs like contraceptives, steroids, metronidazole, anti-retroviral, vaginal drug delivery is the most preferable route. However, achieving sufficient drug concentration in the vagina can be challenging because of its low permeability. The benefits of the vaginal drug delivery system are it increases the bioavailability, least systemic side effects; easiness of use and self-medication is possible. However vaginal drug delivery system is considered as a less effective route because of the unfortunate absorption of drugs across the vaginal epithelium. The traditional commercial preparations, such as creams, foams, gels, irrigations and tablets, are known to reside in the vaginal cavity for a relatively short period of time owing to the self-cleaning action of the vaginal tract and often require multiple daily doses to ensure the desired therapeutic effect. With the rapidly developing field of nanotechnology, the use of specifically designed carrier systems such as Nanoparticle-based drug delivery has been proven an excellent choice for vaginal application to overcome the challenges associated with the low permeability.

Review on formulation and evaluation of solid lipid nanoparticles for vaginal application

In situ gel drug delivery systems are in solution form before administration but once administered, undergo gelation in situ, to form a gel. In the present study nasal in situ gel of Phenylephrine hydrochloride was prepared for the treatment of nasal infections to provide sustained release of drug and to attain site specific action. Carbopol 934 was used as a pH triggered polymer and Poloxamer 188 was used as thermo sensitive agent. Different formulations were prepared by varying the concentrations of Carbopol 934 and Poloxamer 188 polymers in combination with Hydroxylpropyl Methylcellulose (HPMC), HPMC E5LV, HPMC E15LV, and HPMC E50 LV as viscosity enhancing agents. These formulations were evaluated for parameters like drug excipient compatibility, pH, drug content, gelation temperature, viscosity, in vitro drug release, mucoadhesion, ex vivo permeation and stability studies. FTIR study revealed that there was no interaction between drug and polymer. pH of all the formulations were found to be in the range of 5.4-6.2 and the drug content for all the prepared formulations was found to be in the range of 94%-99%. The results of in vitro drug release and mucoadhesive strength indicated that the optimized formulation F15 and F18 is the most successful formulations of the study, exhibited a sustained drug release of in 77.8% in 6 hours and 70.8% in 8 hours with a mucoadhesive strength of 3124.64 and 3167.76 dyne/cm2. From the results it is concluded that Phenylephrine hydrochloride nasal in situ gel produces prolonged and site specific drug delivery for the treatment of respiratory tract infections especially for sinusitis and bronchitis. Antimicrobial and Anti-inflammatory agents are used to treat periodontal diseases. Due to their short half life, these medications have to be taken frequently to maintain the desired therapeutic effect. Extensive efforts have recently been focused on targeting the drugs to a particular region of the body for extended period of time, thus maximizing drug availability and minimizing dose dependent side effects. This article reviews preparation of microspheres, gels and bioadhesive gel incorporated with drug loaded microspheres and parameters to evaluate their efficiency.
