Observations on local spread of pox viruses in tissue.

Das nachstehende Kapitel hat die Aufgabe, demjenigen das Verstandnis der speziellen Kapitel zu erleichtern, der sich mit den Fragen der Infektabwehr noch nicht eingehender befast hat. Es ist nicht moglich, im Rahmen eines solchen informativen Abrisses auf die grundlegenden Erkenntnisse und Theorien erschopfend einzugehen, die gerade auf diesem Gebiete im letzten Jahrzehnt gewonnen wurden. Auswahl und Beschrankung des Stoffes lassen es nicht vermeiden, das die eine oder andere personliche Ansicht des Verfassers starker in Erscheinung tritt, als es bei Berucksichtigung samtlicher Gesichtspunkte der Fall gewesen ware.

Über Resistenz und Immunität

Heute ordnet man vorwiegend auf Grund mikromorphologischer Kriterien in die Gruppe der Pockenvirusarten eine grose Zahl von Erregern menschlicher und tierischer Krankheiten ein, die mannigfache verwandtschaftliche Beziehungen untereinander aufweisen. Die Kenntnis der letzteren ist fur die diagnostische Arbeit im Laboratorium und fur epidemiologische Belange von Bedeutung. Sie ist einerseits die Basis fur die Bemuhungen um eine befriedigende, auch klinischen Bedurfnissen gerecht werdende Pockensystematik, erschwert diese aber andererseits wegen ihrer bisher noch nicht vollstandig erfasten Vielfalt. In Ansehung dieser Schwierigkeit soll hier aus praktischen Erwagungen und nicht prajudizie- rend eine Aufteilung der Pockenviren in 4 Untergruppen vorgenommen werden.

Die Gruppe der quaderförmigen Virusarten (Pockengruppe)

Die Pathologie der Viruskrankheiten ist, wie Nauck (1954) ausfuhrt, z. T. vor grosere Schwierigkeiten gestellt als die Pathologie der bakteriellen Entzundungen. Die Auswertung der im Verlauf einer Virusinfektion auftretenden morphologischen Veranderungen an Zellen und Geweben bereitet erhebliche Schwierigkeiten in der Deutung. Typische oder gar spezifische Reaktionen sind bei den Viruserkrankungen nicht zu erwarten. Viren rufen im Organismus keine spezifischen Reaktionen hervor, sowenig wie das bei bakteriellen Infektionen der Fall ist. Die allgemeinen Probleme der Pathogenese der Virusinfektionen wurden auf S. 456ff. besprochen.

Die Ringzonenbildung auf der mit Geflugelpocken infizierten Chorioallantoismembran des bebruteten huhnereies stellt einen komplexen Vorgang dar, der direkt mit der Virusvermehrung und der lokalen kreisformigen Virusausbreitung im Gewebe zusammenhangt.

An dem komplexen Geschehen beteiligen sich das proliferierte Ektoderm mit konzentrischen Gewebsabschilferungen und das Entoderm mit konzentrischen, wallartigen Gewebsproliferationen. Letztere sind besonders deutlich an der Herdperipherie.

Die Virusspezifitat beider Mechanismen konnte gesichert werden. Ein prinzipieller Unterschied zwischen Primar- und Sekundarherden besteht nicht.

Die lokale Virusausbreitung im Gewebe scheint nicht gleichmasig zu erfolgen. Es wechseln Zonen groser Virusaktivitat und somit starker Gewebs-schadigung mit Zonen geringerer Virusaktivitat und somit schwacherer Gewebsschadigung ab.

Bei der Generalisation siedelte sich das Virus zuerst im Entoderm an und breitete sich von dort weiter aus.

Both the proliferating ectoderm with its concentric exfoliations, and the entoderm with its concentric wall-like proliferations play a part in the process. The latter is particularly prominent at the periphery of the foci.

Both processes are certainly specific for the virus. There is no fundamental difference between primary and secondary foci.

Local virus spread in the tissues seems not to occur uniformly from cell to cell. Zones of greater viral activity and thus more severe cell damage alternate with zones of less activity and less damage.

In the course of generalisation the virus first implants itself in the entoderm and thence spreads to other parts.

— La formation de zones annulaires sur l'allanto-chorion d'oeufs de poule en incubation et infectes par le virus de la variole aviaire est un processus complexe se trouvant en relation directe avec la multiplication et la diffufion circulaire locale du virus dans le tissu.

— Deux feuillets participent a ce processus complexe: l'ectoderme qui a prolifere et presente des desquamations concentriques de tissu et l'endoderme montrant des proliferations tissulaires concentriques et ondulees. Ces dernieres sont particulierement nettes a la peripherie des foyers.

— On put s'assurer que ces deux mecanismes etaient specifiquement dus au virus. Il n'y a aucune difference de principe entre les foyers primaires et les foyers secondaires.

4) 
— La propagation locale du virus dans le tissu ne semble pas se produire d'une facon constante de cellule a cellule. Des zones de grande activite du virus, donc presentant une destruction importante de tissu, alternent avec des zones d'activite moindre montrant des lesions tissulaires moins nettes.

— Dans la generalisation, le virus s'implantait d'abord dans l'endoderme pour se propager ensuite dans d'autres regions.

— La formacion de la zona anular en la membrana corioalantoidea del huevo embrionado de gallina infectado con viruela aviar representa un proceso complejo relacionado directamente con la multiplicacion de los virus y la difusion virica circular local en el tejido.

2°.
— En este hecho complejo participan el ectodermo proliferado con exfoliaciones histicas concentricas y el entodermo con proliferaciones histicas concentricas en forma de rodete. Estas ultimas destacan sobre todo en la periferia focal.

3°.
— Pudo asegurarse la especificidad virica de ambos mecanismos. No existe diferencia marcada entre los focos primarios y secundarios.

4°.
— La difusion virosica local en el tejido no parece realizarse uniformente de celula a celula. Virian zonas de gran actividad viro sica y con ello fuerte lesion histica con zonas de escasa actividad virosica y con ello de mas debil lesion histica.

5°.
— En la generalizacion, el virus se albergaba primero en el entodermo y desde alli se seguia difundiendo.
