For obtaining reliable nanostructural details of large amounts of sample?and if it is applicable?small-angle scattering (SAS) is a prime technique to use. It promises to obtain bulk-scale, statistically sound information on the morphological details of the nanostructure, and has thus led to many a researcher investing their time in it over the last eight decades of development. Due to pressure from scientists requesting more details on increasingly complex nanostructures, as well as the ever improving instrumentation leaving less margin for ambiguity, small-angle scattering methodologies have been evolving at a high pace over the past few decades.As the quality of any results can only be as good as the data that go into these methodologies, the improvements in data collection and all imaginable data correction steps are reviewed here. This work is intended to provide a comprehensive overview of all data corrections, to aid the small-angle scatterer to decide which are relevant for their measurement and how these corrections are performed. Clear mathematical descriptions of the corrections are provided where feasible. Furthermore, as no quality data exist without a decent estimate of their precision, the error estimation and propagation through all these steps are provided alongside the corrections. With these data corrections, the collected small-angle scattering pattern can be made of the highest standard, allowing for authoritative nanostructural characterization through its analysis. A brief background of small-angle scattering, the instrumentation developments over the years, and pitfalls that may be encountered upon data interpretation are provided as well.

Everything SAXS: small-angle scattering pattern collection and correction

Beamline 4-2 at the Stanford Synchrotron Radiation Laboratory is a small-angle X-ray scattering/diffraction facility dedicated to structural studies on mostly noncrystalline biological systems. The instrument consists of a pinhole camera, which covers the magnitude of the scattering vector Q in the range 0.004–1.3 A−1 [Q = (4π/λ)sin θ, where θ and λ are one half of the scattering angle and the X-ray wavelength, respectively], and a Bonse–Hart geometry ultra-small-angle X-ray scattering setup for the Q range an order of magnitude smaller. The pinhole camera allows quick automated distance and detector selection among any combination of five distances and three position-sensitive detectors. The double-crystal monochromator can have either Si 111 crystals or a pair of synthetic multilayer diffractive elements for higher flux applications. We have adopted a suite of software originally developed for macromolecular crystallography for integrated beamline control as well as static and slow time-resolved small-angle scattering data collection. This article outlines recent technological developments and specialized instrumentation for conducting noncrystalline scattering experiments in structural biology at improved time and spatial resolutions.

Biological small-angle X-ray scattering facility at the Stanford Synchrotron Radiation Laboratory

A Bonse–Hart camera suitable for studying dilute and weakly scattering systems has been developed. The required level of reduction of the parasitic background scattering was realised by replacing channel-cut crystals by a pair of polished and deeply etched parallel crystals. The performance of this ultra-small-angle X-ray scattering instrument is illustrated by means of two examples: the microstructure of a short-range attractive colloidal system; and the growth dynamics of soot particles in a diffusion flame. In addition, the feasibility of performing dynamic X-ray scattering experiments is demonstrated.

Everything SAXS: Small-angle scattering pattern collection and correction

The formation of precursors and the growth and aggregation of silicalite-1 crystals using tetrapropylammonium as a template (Si-TPA-MFI) has been studied in situ using X-ray scattering techniques. Simultaneous small-angle and wide-angle X-ray scattering (respectively SAXS and WAXS) experiments using synchrotron light showed that the formation of amorphous colloidal aggregates in water clear synthesis mixtures was dependent on the alkalinity of the solutions. In situ time-resolved ultra-small-angle X-ray scattering (USAXS) showed the form factor oscillations of the growing crystals. Fitting of the USAXS patterns to the scattering pattern of spherical particles having a normal particle size distribution showed a linear growth of the average crystal diameter, which was approximately the same for both alkalinities studied. The final size of the crystals was highest for the synthesis mixture having the highest alkalinity, which can be explained in terms of number of viable nuclei formed. At the end of the line...

In Situ Investigation of Si-TPA-MFI Crystallization Using (Ultra-) Small- and Wide-Angle X-ray Scattering

We report on first experiments carried out on the high brilliance undulator beamline (BL4) at the ESRF. A Bonse-Hart camera was installed in order to examine the performance of the X-ray beam and its source for this type of experiments. The results show that excellent data can be obtained very fast. They belong to a scattering vector range which is partially covered by visible light scattering experiments. The limiting problem is caused by diffuse scattering from the channel-cut crystals (up to 5 reflections) but according to our results it should be possible to decrease this effect substantially by using a still higher number of reflections while still maintaining unprecedentedly high count rates and dynamic range due to the very bright source.
