Including appendixes, this book contains 894 pages of text. The entire panorama of the neural sciences is surveyed and examined, and it is comprehensive in its scope, from genomes to social behaviors. The editors explicitly state that the book is designed as "an introductory text for students of biology, behavior, and medicine," but it is hard to imagine any audience, interested in any fragment of neuroscience at any level of sophistication, that would not enjoy this book. Sprague Dawley rats weighing 225-250 g were used for evaluating wound healing and gastroprotective activities. The plant extracts of J.sambac and H.colorata were assessed in-vivo for toxicity study. Antioxidant properties of the crude extracts were determined using DPPH radical scavenging assay and total phenolic content. Β-sitosterol and octyl gallate compounds were isolated from J. sambac and H. Colorata petroleum ether crude extracts by CC, TLC and HPLC and were evaluated for their gastroprotective activities.Macroscopic finding showed that H.colorata and J.sambac significantly accelerated wound healing compared to vehicle. Post wounding tissues collected on day 10th for oxidative damage evaluation revealed significant increase in antioxidant enzymes (SOD and GPx) and decrease in the MDA level. Grossly, and histology showed smooth surfaces, marked reduction of edema and leucocytes infiltration of submucosal layer compared with ulcer group. Immunohistochemistry revealed overexpression of Hsp70 protein and downexpression of Bax protein in rats. Significant increase in the pH, mucus of gastric content and gastric PGE2, SOD, COX -2 and reduced level of MDA were observed. Β-sitosterol compound showed significant gastroprotective activity against acidified ethanol-induced gastric damage in rats more than Octyl gallate compound. Moreover, β-sitosterol recorded significant increase in the pH, gastric mucus and decrease in the ulcer area. Biochemistry, histopathology and immunohistochemistry results revealed significant improvement in β-stosterol than Octyl gallate.Wound healing can be accelerated and gastric ulcer can be inhibited by the antioxidants activity of H.colorata and J.sambac leaves extracts and its active antioxidant ingradient β-sitosterol while the normal gastric status can be preserved.

Cu/Zn Superoxide Dismutase (SOD1) plays a critical role in alleviating oxidative stress by catalyzing the conversion of superoxide radicals into oxygen and hydrogen peroxide. This review presents an in-depth analysis of the challenges and strategies involved in optimizing SOD1 production, with a focus on Pichia pastoris as an expression system. Key approaches such as the strategic selection of expression vectors, codon optimization, and the fine-tuning of fermentation parameters to maximize SOD1 yield are thoroughly explored. Advances in protein engineering, the co-expression of molecular chaperones, and the use of stabilizers and additives are also examined for their role in improving SOD1 stability and functionality. The review highlights the significant biomedical and industrial applications of overexpressed and thermostable SOD1, uncovering novel opportunities for therapeutic interventions and biotechnological innovations. Advancing Cu/Zn Superoxide Dismutase (SOD1) production in Pichia pastoris : challenges, strategies, current research status, and future directions

Charcot-Marie-Tooth disease (CMT) 2F and distal Hereditary Motor Neuropathy (dHMN) are peripheral motor axonopathies with limited sensory involvement, which usually present during the first decade of life. They are caused by mutations in heat shock protein 27 (Hsp27)/HSPB1, a highly conserved, ubiquitously expressed molecular chaperone. Hsp27 has several cytoprotective functions including the inhibition of apoptosis, protection against oxidative stress and promotion of axonal growth. In this Thesis, the effects of several pathogenic Hsp27 mutations were examined to elucidate their cellular effects in vitro and map these effects to different regions of the gene.
The effects of Hsp27 mutations were first investigated in neuronal-like SH-SY5Y cells in vitro Analysis of cell survival and cellular morphology revealed that all mutations were cytotoxic under basal conditions. However, mutations located in the α-crystallin protein domain of Hsp27 resulted in a significant increase in the vulnerability of cells to cytoskeletal stressors and decreased neurite outgrowth.
Using immunocytochemistry, interactions between mutant Hsp27 and cytoskeletal components were also examined. Mutations located in the Hsp27 α-crystallin domain increased co-localisation of Hsp27 with cytoskeletal elements. Although the mutation within the N-terminus did not have this effect it did result in the formation of distinct nuclear aggregates containing mutant Hsp27.
The functional effects of Hsp27 mutations were investigated using lentiviral delivery of mutant Hsp27 in primary motoneurons. Examination of mitochondrial function showed that none of the Hsp27 mutants had any effect on mitochondrial membrane potential.
The results presented in this Thesis show that disease-causing Hsp27 mutations have differential effects upon protein function in vitro depending upon the gene position of the mutation. Therefore, although all Hsp27 mutations in CMT patients result in motoneuron degeneration, these results suggest that this process may be initiated by different pathological mechanisms and that normal Hsp27 function is essential for the maintenance of motor-axonal function.
