Brassica napus is an economically important crop across different continents including temperate and subtropical regions in Europe, Canada, South Asia, China and Australia. Its widespread cultivation also brings setbacks as it plays host to fungal, oomycete and chytrid pathogens that can lead to serious yield loss. For sustainable crop production, identification of resistance (R) genes in B. napus has become of critical importance. In this review, we discuss four key pathogens affecting Brassica crops: Clubroot (Plasmodiophora brassicae), Blackleg (Leptosphaeria maculans and L. biglobosa), Sclerotinia Stem Rot (Sclerotinia sclerotiorum), and Downy Mildew (Hyaloperonospora parasitica). We first review current studies covering prevalence of these pathogens on Brassica crops and highlight the R genes and QTL that have been identified from Brassica species against these pathogens. Insights into the relationships between the pathogen and its Brassica host, the unique host resistance mechanisms and how these affect resistance outcomes is also presented. We discuss challenges in identification and deployment of R genes in B. napus in relation to highly specific genetic interactions between host subpopulations and pathogen pathotypes and emphasize the need for common or shared techniques and research materials or tighter collaboration between researchers to reconcile the inconsistencies in the research outcomes. Using current genomics tools, we provide examples of how characterization and cloning of R genes in B. napus can be carried out more effectively. Lastly, we put forward strategies to breed resistant cultivars through introgressions supported by genomic approaches and suggest prospects that can be implemented in the future for a better, pathogen-resistant B. napus.

Current Status and Challenges in Identifying Disease Resistance Genes in Brassica napus

The genus Brassica contains a wide range of diploid and amphipolyploid species including some of the most important vegetable, condiment and oilseed crops worldwide. As members of the Brassicaceae family the brassicas are the closest crop relatives to the model plant Arabidopsis thaliana, and hence are major beneficiaries from the vast array of Arabidopsis molecular genetic and genomic tools and the increasingly good annotation to major Brassica crop genomes. In this review examples are shown from recent studies that demonstrate the potential for intergenome navigation from model to crop plant and for comparisons among genetic and cytogenetic maps between the model and crop species and among different crop brassicas. The use of interspecific and intergeneric hybridization for introgression of novel traits into Brassica genomes from the secondary and tertiary crucifer genepools is described. In this context the use of the Brassica triangle of three diploid species and their corresponding amphiploids as an excellent model system for studying the mechanisms and control of homeologous recombination and polyploidization is discussed from a crop breeding perspective.

Cytogenetics and genome analysis in Brassica crops

The infection by Leptosphaeria maculans of Brassica napus cultivars with major gene resistance derived from Brassica rapa subsp. sylvestris was studied in southeastern Australia. Following the commercial release of these cultivars in Australia in 2000, plants with stem cankers were first reported in 2002 at two geographically isolated regions in South Australia and New South Wales. In 2003, this study showed that the major gene resistance had been overcome in an area of approximately 50,000 ha in South Australia and in two fields in New South Wales (0.5 and 30 ha). There was no relationship between disease severity and incidence in 2003 and the proximity to the sites where resistance breakdown occurred in 2002. At some locations, the frequency of isolates able to overcome the B. rapa subsp. sylvestris-derived resistance had increased between 2002 and 2003. Isolates cultured from canola cultivars with either B. rapa subsp. sylvestris-derived resistance or polygenic resistance showed host specificity when inoculated onto cultivars with B. rapa subsp. sylvestris-derived or polygenic resistance, respectively. The most likely cause of the resistance breakdown was the rapid increase in frequency of L. maculans isolates virulent on this particular resistance source. The selection pressure leading to this increased frequency was probably mediated by the planting of cultivars harboring the major resistance gene in the same locations for a 3-year period, and the ability of the pathogen to produce large numbers of asexual and sexual spores.

Major Gene Resistance to Blackleg in Brassica napus Overcome Within Three Years of Commercial Production in Southeastern Australia.

Interspecific hybridization is widespread in nature, where it can lead to either the production of new species or to the introgression of useful adaptive traits between species. In agricultural systems, there is also great potential to take advantage of this process for targeted crop improvement. In the Brassica genus, several crop species share close relationships: rapeseed (Brassica napus) is an ancestral hybrid between turnip (B. rapa) and cabbage (B. oleracea), and mustard species B. juncea, B. carinata and B. nigra share genomes in common. This close relationship, plus the abundance of wild relatives and minor crop species in the wider Brassiceae tribe which readily hybridize with the Brassica crop species, makes this genus an interesting example of the use of interspecific hybridization for crop improvement. In this review we introduce the Brassica crop species and their wild relatives, barriers to interspecific and intergeneric hybridization and methods to overcome them, summarize previous successful and unsuccessful attempts at the use of interspecific hybridization for crop improvement in Brassica, and provide information about resources available to breeders wishing to take advantage of this method in the Brassica genus.

Interspecific Hybridization for Brassica Crop Improvement

Somatic hybrids between Brassica napus (oilseed rape) and its wild relative Sinapis arvensis (Xinjiang wild mustard) from northwestern China were produced by fusing mesophyll protoplasts. Fifty-four plants were identified as symmetric hybrids and four as asymmetric hybrids by random amplified polymorphic DNA analysis and nuclear DNA content. The morphology of investigated 58 hybrid plants resembled characters from both parental species. Highly fertile hybrids were recovered where the fertility was associated with the choice of B. napus genotype. Enhanced disease resistance to Leptosphaeria maculans was found in S. arvensis and in the hybrid offspring. This plant material has great potentials not only for use as a bridge for the introduction of a number of valuable traits from the wild species to Brassica crops but also for breeding new varieties with improved blackleg resistance.

Production of fertile intergeneric somatic hybrids between Brassica napus and Sinapis arvensis for the enrichment of the rapeseed gene pool

Blackleg caused by Leptosphaeria maculans is one of the most important diseases affecting oilseed rape worldwide. Sinapis arvensis is valuable for the transfer of blackleg resistance to oilseed rape (Brassica napus) because this species contains high resistance against various aggressive isolates of the blackleg fungus. These include at least one Australian isolate which has been found to overcome resistance originating from species with the Brassica B genome, until now the major source for interspecific transfer of blackleg resistance. Backcross offspring from intergeneric crosses between Brassica napus and S. arvensis were subjected to phytopathological studies and molecular cytogenetic analysis with genomic in situ hybridisation (GISH). The BC3S progenies included fertile plants exhibiting high seedling (cotyledon) and adult plant resistance associated with the presence of an acrocentric addition chromosome from S. arvensis. In addition, some individuals with adult plant resistance but cotyledon susceptibility were observed to have a normal B. napus karyotype with no visible GISH signals, indicating possible resistant introgression lines. Phytopathological analysis of selfing progenies from 3 different highly resistant BC3 plants showed that seedling and adult plant resistance are probably conferred by different loci.
