The reaction to Tomato spotted wilt virus (TSWV) was evaluated in 94 accessions of Nicotiana, originating from the Institute of Soil Science and Plant Cultivation tobacco germplasm collection in Pulawy, Poland. Tests for resistance were conducted under greenhouse conditions using single TSWV isolate collected from tobacco plantation in Lublin district, Poland. The presence of the virus was verified using DAS-ELISA. SCAR markers associated with TSWV resistance gene were applied. The members of the section Alatae, the genus Nicotiana: N. alata, N. forgetiana, and Nicotiana x sanderae as well as N. tabacum cultivars: ‘Polalta’ and ‘Wiktoria’ with the TSWV resistance gene introduced from N. alata, displayed the hypersensitive reaction (HR) against TSWV (grade 0 on symptom intensity scale). In some of those accessions, the virus spread from the initially infected areas eliciting systemic hypersensitive reaction (SHR). Five accessions of N. alata and three of Nicotiana

 sanderae were composed of 6.3–50.0 % of plants in which SHR symptoms appeared. In all of N. forgetiana plants HR reaction was followed by systemic infection (SHR). In N. tabacum ‘Wiktoria’ 21.1 % of plants showed HR reaction, while the remaining were susceptible (S). All of the genotypes which responded with HR or SHR reaction to TSWV infection demonstrated the presence of SCAR markers linked to the resistance gene. The remaining eighty tested accessions were identified as being susceptible upon exposure to TSWV.

A survey of Nicotiana germplasm for resistance to Tomato spotted wilt virus (TSWV)

There are more than 80 naturally occurring relatives of cultivated tobacco (Nicotiana tabacum L.) in the genus Nicotiana. In this review, we examine how and to what extent these natural germplasm resources have been utilized in hybridization and introgression experiments over the past century. To date, more than 400 interspecific Nicotiana hybrids have been reported. We focus on individual Nicotiana species involved in interspecific hybrids with cultivated tobacco produced by sexual and asexual methods, including the recently discovered grafting method. Problems related to the hybridization of N. tabacum with other species, namely, cross-incompatibility, maternal phenotypes in hybrid offspring, interspecific incongruity, lethality of juvenile hybrids, and sterility of viable hybrids, are reviewed. Among the 58 interspecific hybrids involving N. tabacum reported thus far, 25 were also reported as somatic hybrids and two were obtained only as somatic hybrids. Thirty-six sterile sexual F1 hybrids were converted to fertile or partly fertile allopolyploids. Sixteen Nicotiana species have been deployed as a source of usable traits that were introgressed into N. tabacum, offering resistance to or tolerance of pathogens or pests. The mechanisms of introgression, such as alien addition and substitution, as well as the barriers and limitations of introgression, including erratic inheritance and adverse linkages, are discussed. Thirty-one Nicotiana species used as sources of cytoplasmic male sterility in N. tabacum have produced multiple alloplasmics; most showed negative effects of alien cytoplasm but a few have been deployed successfully in hybrid cultivars of N. tabacum.

The Use of Nicotiana Species in Tobacco Improvement

Transgenesis allows to modify plant genomes relatively quickly and precisely. Moreover, comparing with classical methods genetic transformation significantly facilitates and shortens the time needed for breeding a new cultivar. Shortening a breeding process is possible thanks to obtaining haploids and then doubled haploids being homozygotes as regards all traits which increase the efficiency of selection of desired recombinants. The plant material used in the study were transgenic tobacco doubled haploids containing Lettuce mosaic virus coat protein gene (LMV CP), which determines resistance to Potato virus Y (PVY). The doubled haploids (DH) were previously obtained from hybrids coming from crossing transgenic cultivar MN 944 with cv. Wiślica. DH plants were obtained by combination of anther culture and spontaneous doubling number of chromosomes under in vitro conditions. In the present study, the generation DH 2 of doubled haploid lines obtained by self-pollination were estimated by comparison with their parental cultivars. The experiment was conducted under greenhouse conditions. The evaluation of agronomic traits included elements of growth and development such as growth rate, number of days to flowering, area of leaves, height and morphology of plants. Cured leaves of DH lines were analyzed as regards selected physical and chemical traits. Individual DH lines differed among themselves and in comparison to parental cultivars as regards some traits. However, only little diversity within the lines was noticed. The DH lines characterized by the most satisfying agronomic traits may be valuable initial material in further breeding.

Agronomic characteristics of transgenic tobacco doubled haploids resistant to Potato virus Y

Thielaviopsis basicola and Tomato spotted wilt virus (TSWV) are the most important problems in a moderate climate zone. Previously obtained doubled haploids (DH) of F1 hybrids of the flue-cured line WGL3 resistant to Th. basicola and the dark-cured line PW-834 carrying RTSW-al gene provided the research material. Biological tests and SCAR markers linked with TSWV were applied to confirm resistance of DH. Lines combining resistance to TSWV and Th. basicola were evaluated for morphological and chemical characteristics. Most of DH were significantly shorter than parents but two lines, 31/A/2 and 31/B/3, were close to the flue-cured WGL3. Usually DH possessed fewer leaves while one of them 31/B/3, exceeded parental forms. The doubled haploids flowered later than their parents. The most negative effect was reduced area of mid-position leaves of DH. It might be explained by a recombination during microsporogenesis in F1, however the influence of 'Polalta'-derived RTSW-al gene cannot be excluded. Extensive line to line variation for nicotine and sugars content was not associated with the genes for TSWV and Th. basicola resistance. Biological tests and field performance of DH revealed potential to overcome the negative effect of coupling between the RSTV-al gene and genes responsible for the morphological deformations.

Morphological and chemical characteristics of doubled haploids of flue-cured tobacco combining resistance to Thielaviopsis basicola and TSWV

Black root rot (BRR) caused by Thielaviopsis basicola as well as Tomato spotted wilt virus (TSWV) are the most serious problems in tobacco growing regions. We crossed the breeding line WGL 3 carrying BRR resistance derived from N.glauca with the line PW-834 the resistance of which to TSWV was transferred from cultivar Polalta. Anthers obtained from F1 hybrid plants were cultured to induce haploids combining resistance to Th. basicola and TSWV. Flow cytometry analysis revealed 242 haploids and 2 spontaneous doubled haploids among regenerants. All haploids were cloned and then evaluated for BRR as well as TSWV resistance. The presence of pathogens was detected by microscopic evaluation of roots or using DAS-ELISA test. Microscopic assessment showed that, 132 haploids had no symptoms of Th. basicola which, together with the absence of symptoms in the F1 hybrids, indicated a dominant monogenic mode of inheritance. At the same time only 30 haploids demonstrated resistance to TSWV. SCAR markers associated with TSWV resistance gene detection was applied. The results indicate that small proportion of TSWV-resistant haploids is probably due to the influence of deleterious genes flanking the resistance factor that reduced vitality of gametophytes. Altogether, 24 haploids showed multiple resistance to Th. basicola and TSWV.

Abstract

Nicotiana tabacum cv. ‘Polalta’ is reported as having a hypersensitive type of Tomato spotted wilt virus (TSWV) resistance. In ‘Polalta’ hybrids with susceptible cultivars, morphological deformations (thickened and ribbon-shaped leaves, irregular venation, tumours) appear. Acceptable DH lines derived from hybrids of ‘Polalta’ with ‘Wiślica’ were obtained by a method of in vitro culture. Those DH lines were evaluated for TSWV resistance. Tests for resistance were conducted under greenhouse conditions using one TSWV isolate from a local source. The presence of the virus was verified using DAS-ELISA. The DH lines PW-833, PW-834, and PW-900 showed a near correct morphology and stably inherited dominant monogenic resistance to TSWV.
